Indication

Bolemide (Permalidomide) is a thalidomide analog in combination with dexamethasone for patients with multiple myeloma who have received at least two prior therapies including lenalidomide and a proteasome inhibitor , and who have been Disease progression was confirmed within 60 days of completion of the last treatment .

Dosage and Administration
1. Administration

 Multiple myeloma : pomalidomide 4 mg orally per day on days 1-21 of a cycle  of 28 days ;

 In combination with 40 mg daily on days 1 , 8 , 15, and 22 of each cycle  more than 

For 75-year-olds, the initial dose of dexamethasone is daily on days 1 , 8, 15, and 22 of each cycle. 

2 0mg each time. Cycles are repeated until disease progression or toxicity is not tolerated.

2. Dosage Form Specifications

●  Capsules, 2 mg, 3 mg, 4 mg. 3. Contraindications: Pregnant women are prohibited .

Warnings and Precautions

 ●  Hematologic toxicity : The most common grade 3/4  adverse  event was neutropenia . Monitor patients for hematologic toxicity , especially neutropenia .

•  Hepatotoxicity: Liver failure including fatal events; monitor liver function tests monthly .

●  Hypersensitivity reactions : Angioedema and severe dermatological reactions have been reported . vascular water

Swelling and severe dermatological reactions discontinue pomalidomide therapy .

Tumor  lysis syndrome (TLS ) : monitor patients  at risk for TLS (eg , patients with high tumor burden) 

) and take appropriate precautions .

Adverse reactions

Most common adverse reactions (  30%) : fatigue, asthenia , neutropenia , anemia , constipation , nausea , diarrhea , dyspnea, upper respiratory tract infection , back pain and fever.
